Extreme homogeneity among Brazilian wheat genotypes determined by RAPD markers PAB
FREITAS,LORETA BRANDÃO DE; JERUSALINSKY,LEANDRO; BONATTO,SANDRO LUIS; SALZANO,FRANCISCO MAURO.
Random amplified polymorphic DNA markers (RAPD) were used to estimate the variability of 14 genotypes of Brazilian wheat (Triticum aestivum L.), using a set of 50 random 10mer primers. A total of 256 reproducibly scorable DNA amplification products were obtained from 48 of the primers, 83% of which were polymorphic. Genetic distances among genotypes were calculated and a dendrogram and a principal coordinates analysis showing the genetic relationships among them were obtained. Despite the low variability found (average genetic distance of 27%), two groups of genotypes could be identified, which probably reflect how they were formed. Studies such as this one may be important in the planning and development of future improvement programs for this plant...

Genetic distance between broodstocks of the marine shrimp Litopenaeus vannamei (Decapoda, Penaeidae) by mtDNA analyses Genet. Mol. Biol.
Francisco,Ana Karina de; Galetti Junior,Pedro Manoel.
In the constantly growing Brazilian shrimp industry the evaluation of genetic relationships between broodstocks is an useful tool for shrimp culture management programs. We established the genetic relationships between five broodstocks of the white marine shrimp Litopenaeus vannamei (Penaeidae) based on the sequencing of the mtDNA 16S rRNA and cytochrome oxidase I (COI) regions. Although no divergence was found between the broodstocks for the highly conservative 16S rRNA gene, we did find an 8.2% distance between L. vannamei and the Farfantepenaeus subtilis. Analyses of the COI region showed genetic distances of only 0.2 to 1% between the broodstocks, which contrasted with the 10.9% mean distance found between L. vannamei and F. subtilis. The small genetic...

Genetic diversity between native and improved Cattleya walkeriana Gardner famous clones Agronomy
Tambarussi, Evandro Vagner; Ann Veasey, Elizabeth; Menezes, Lou; Ibañes, Bruna; Lombardi, Kátia Cylene; Vencovsky, Roland.
The aim of this study was to evaluate the genetic diversity among native plants and some individuals obtained from crosses with unknown genealogy of C. walkeriana as well as C. loddigesii and C. nobilior and to advance towards solving the question of the genetic purity of the “Orchidglade” clone. Eight microsatellite loci were used to evaluate the genetic diversity between individuals of C. walkeriana. Microsatellites were not efficient in determining the genetic diversity between C. walkeriana groups (native and improved). The difficulty in determining the genetic distance between the different genotypes can be attributed to the complex mating system of the species and to a weak genetic barrier that facilitates the development of hybrids. Our analysis...

Philippsen,Juliana S.; Renesto,Erasmo; Gealh,Ana Maria; Artoni,Roberto F.; Shibatta,Oscar A.; Zawadzki,Claudio H..
Four samples of Neoplecostomus yapo were analyzed through the allozyme electrophoresis technique in corn starch gel. The allozyme pattern was similar to those found in N. paranensis with 24 loci scored. Two samples (ribeirão Atlântico and ribeirão Uraí) showed monomorphic bands for all 24 loci, whereas the other two (rio Verde and rio Fortaleza) showed 8.3% of polymorphic loci. The He genetic variability estimates for the rios Verde and Fortaleza populations were 0.0195 and 0.0179, respectively, too much inferior to the mean heterozygosity summed to species from the whole world (0.051). The Wright statistical values F IS = 0.5181, F IT = 0.5681 and F ST = 0.1039 and the genetic distance of Nei values showed that the four samples are genetically very...

Genetic variability in the Skyros pony and its relationship with other Greek and foreign horse breeds Genet. Mol. Biol.
Bömcke,Elisabeth; Gengler,Nicolas; Cothran,E. Gus.
In Greece, seven native horse breeds have been identified so far. Among these, the Skyros pony is outstanding through having a distinct phenotype. In the present study, the aim was to assess genetic diversity in this breed, by using different types of genetic loci and available genealogical information. Its relationships with the other Greek, as well as foreign, domestic breeds were also investigated. Through microsatellite and pedigree analysis it appeared that the Skyros presented a similar level of genetic diversity to the other European breeds. Nevertheless, comparisons between DNA-based and pedigree-based results revealed that a loss of genetic diversity had probably already occurred before the beginning of breed registration. Tests indicated the...

Genetic variation among and within tritipyrum (Thinopyrum bessarabicum x Triticum durum) lines using PCR-based molecular markers Ciencia e Investigación Agraria
Siahsar,Barat Ali; Allahdoo,Maryam; Shahsevand Hassani,Hossein.
Primary hexaploid tritipyrum lines are amphiploids between Triticum durum and Thinopyrum bessarabicum, which can set seed in at least 250 mM NaCl. Genetic variation within and among 14 lines of primary tritipyrum were investigated by using 2 random and 6 semi random primers that produced clear and reproducible DNA amplicons. Average values for Nei's gene diversity (h) and Shannon indices (I) were 0.115 and 0.178, respectively. These results indicated low variation within primary tritipyrum lines, probably related to unbiased chromosome and partial aneuploidy. Total genetic diversity (Ht) and gene diversity within lines (Hs) were 0.2114 and 0.1156, respectively. The coefficient of genetic differentiation (Gst) was equal to 0.4534. AMOVA showed highly...

Genetic variation among species, races, forms and inbred lines of lac insects belonging to the genus Kerria (Homoptera, Tachardiidae) Genet. Mol. Biol.
Ranjan,Sanjeev Kumar; Mallick,Chandana Basu; Saha,Dipnarayan; Vidyarthi,Ambarish S; Ramani,Ranganathan.
The lac insects (Homoptera: Tachardiidae), belonging to the genus Kerria, are commercially exploited for the production of lac. Kerria lacca is the most commonly used species in India. RAPD markers were used for assessing genetic variation in forty-eight lines of Kerria, especially among geographic races, infrasubspecific forms, cultivated lines, inbred lines, etc., of K. lacca. In the 48 lines studied, the 26 RAPD primers generated 173 loci, showing 97.7% polymorphism. By using neighbor-joining, the dendrogram generated from the similarity matrix resolved the lines into basically two clusters and outgroups. The major cluster, comprising 32 lines, included mainly cultivated lines of the rangeeni form, geographic races and inbred lines of K. lacca. The...

Genetic variation and trade-offs for reproduction ans survival in the Pacific oyster Crassostrea gigas ArchiMer
Boudry, Pierre; Degremont, Lionel; Bedier, Edouard; Pouvreau, Stephane; Normand, Julien; Ernande, Bruno.
To date, the most significant method to genetically reduce reproductive effort in the Pacific oyster (Crassostrea gigas) has been through the production of triploids, especially since the development of tetraploid lines allowing the breeding of 'natural' triploids. Gametogenesis of triploid oysters is strongly reduced compared with diploids, although they are not fully sterile and can produce viable gametes and some progenies when crossed with diploids. Reduced reproductive allocation and higher heterozygocity are commonly proposed as the main reasons why triploids often present superior yield compared with diploids. Temperature and food availability are known to favour gametogenesis in triploids, but the existence of genetic variation for this trait...

Genetic variation in cultivated Rheum tanguticum populations Genet. Mol. Biol.
Hu,Yanping; Xie,Xiaolong; Wang,Li; Zhang,Huaigang; Yang,Jian; Li,Yi.
To examine whether cultivation reduced genetic variation in the important Chinese medicinal plant Rheum tanguticum, the levels and distribution of genetic variation were investigated using ISSR markers. Fifty-eight R. tanguticum individuals from five cultivated populations were studied. Thirteen primers were used and a total of 320 DNA bands were scored. High levels of genetic diversity were detected in cultivated R. tanguticum (PPB = 82.19, H = 0.2498, H B = 0.3231, I = 0.3812) and could be explained by the outcrossing system, as well as long-lived and human-mediated seed exchanges. Analysis of molecular variance (AMOVA) showed that more genetic variation was found within populations (76.1%) than among them (23.9%). This was supported by the coefficient...

Genetic variation of wild and hatchery populations of the catla Indian major carp (Catla catla Hamilton 1822: Cypriniformes, Cyprinidae) revealed by RAPD markers Genet. Mol. Biol.
Rahman,S.M. Zakiur; Khan,Mukhlesur Rahman; Islam,Shahidul; Alam,Samsul.
Genetic variation is a key component for improving a stock through selective breeding programs. Randomly amplified polymorphic DNA (RAPD) markers were used to assess genetic variation in three wild population of the catla carp (Catla catla Hamilton 1822) in the Halda, Jamuna and Padma rivers and one hatchery population in Bangladesh. Five decamer random primers were used to amplify RAPD markers from 30 fish from each population. Thirty of the 55 scorable bands were polymorphic, indicating some degree of genetic variation in all the populations. The proportion of polymorphic loci and gene diversity values reflected a relatively higher level of genetic variation in the Halda population. Sixteen of the 30 polymorphic loci showed a significant (p < 0.05, p...

Genetic variations of EBV-LMP1 from nasopharyngeal carcinoma biopsies: potential loss of T cell epitopes BJMBR
Tang,Y.L.; Lu,J.H.; Cao,L.; Wu,M.H.; Peng,S.P.; Zhou,H.D.; Huang,C.; Yang,Y.X.; Zhou,Y.H.; Chen,Q.; Li,X.L.; Zhou,M.; Li,G.Y..
To find Epstein-Barr virus (EBV) strains with genetic variations of EBV latent membrane protein 1 (EBV-LMP1) from nasopharyngeal carcinoma (NPC), the full-length DNA of LMP1 genes from 21 NPC biopsies obtained in Hunan province in southern China was amplified and sequenced. Our sequences were compared to those previously reported by the Clustal V method. Results showed that all 21 sequences displayed two amino acid changes most frequently in LMP1 of CD4+ T cell epitopes at codons 144 (F®I, 21/21) and 212 (G®S, 19/21) or (G®N, 2/21). We also show that type A EBV strain is prevalent in the cases of NPC from Hunan province with a 30-bp 18/21 deletion, and we highlight that this deletion resulted in loss of one of the CD4+ T cell-restricted epitopes. The other...
